Ferroptosis is a form of regulated cell death driven by iron and the uncontrolled oxidation of fat molecules in cell membranes. Unlike apoptosis, which is a neat and programmed dismantling of a cell, ferroptosis involves a runaway chemical reaction called lipid peroxidation that damages membranes and quickly kills the cell. Cells normally protect themselves from this by using antioxidant systems, especially glutathione and the enzyme glutathione peroxidase 4 (GPX4), to neutralize harmful lipid peroxides. When iron levels are high or antioxidant defenses are weakened, those protective systems fail and ferroptosis can occur. Researchers identify ferroptosis by biochemical signs like increased lipid peroxides, sensitivity to iron chelators, and dependence on specific metabolic pathways. Ferroptosis matters because it plays a role in many health conditions where cells are stressed or injured, including some neurodegenerative diseases, organ damage after loss of blood flow, and certain types of cancer. In some diseases, unwanted ferroptosis can make tissue damage worse; in others, inducing ferroptosis can be a way to kill stubborn cancer cells that resist other treatments. Because it uses distinct chemistry from other forms of cell death, scientists are developing drugs that either block ferroptosis to protect tissues or trigger it to eliminate harmful cells. Understanding ferroptosis helps explain how cells die under specific conditions and opens new avenues for therapies and protective strategies.
